The pigment production characteristics of the bacterium under various conditions were studied. The optimal agar plate ingredients for pigment production by the bacterium were first studied: the optimal ingredients were 5 g/liter glucose, 10 g/liter tryptic soy broth, and 40 g/liter glycerol at pH 6.4. Bacterial cells grew on the agar plate during the incubation, while the pigment spread into the agar plate, meaning that it is water soluble.
Indigoidine is a natural pigment formed by the condensation of two L-glutamine molecules catalyzed by a non-ribosomal peptide synthetase (NRPS). It is characterized by a very distinctive blue coloration, similar to indigo, from which it gets its name (Figure 2b). This compound is triggered in bacteria by previously known biosynthetic gene clusters.
It is estimated that indigoidine provides the strain that produces it with an increase in resistance to oxidative stress, acting as an antioxidant, protecting the bacterium from reactive oxygen species [7,34]. Also, there are reports that indigoidine has antibacterial activity against Vibrio fischeri (Table 1), inhibiting its growth [36]. Actual studies have concentrated mainly on understanding the pigment’s usefulness and function.
The blue color of the indigoidine compound is bright and sustainable. As a natural dye, it has promising health benefits as an antioxidant and antimicrobial actor. Because of a new purification process patented with the compound, indigoidine natural blue dye is safe to use in food and drinks.
The indigoidine compound developed at Utah State uses an innovative method to produce a natural, safe deep blue dye that retains its brightness. Indigoidine natural blue dye is a promising alternative to the synthetic dyes used to colored jeans, leather, food, beverages, cosmetics and paper.
